The kit is based on principle of Sandwich-CLIA. The micro CLIA plate of the kit has been pre-coated with an antibody specific to Mouse NOS3/eNOS. test sample and standards are added to the micro CLIA plate wells and combined with the specific antibody. Then a detection antibody (biotinylated) specific for Mouse NOS3/eNOS and Avidin-Horseradish Peroxidase (HRP) conjugate are added to each well of the well and then the plate is incubated for specific time. The free conjugate is washed away by buffer. The substrate solution is added to each well. The well that contains Mouse NOS3/eNOS, detection antibody (biotinylated) and Avidin-HRP conjugate will appear luminescence. At 420nm, the Relative light unit (RLU) value is measured by the Chemiluminescence immunoassay analyzer. The obtained RLU value is positively associated with the concentration of Mouse NOS3/eNOS. The concentration is calculated for Mouse NOS3/eNOS in the samples by comparing the RLU value of the samples with the standard curve.
