The kit is based on principle of Competitive-ELISA. The kit contains micro ELISA plate coated with Universal GSH. The chemical reaction, Universal GSH available in test sample or standard competes with a fixed amount of Universal GSH on the solid phase of the plate sites on the detection antibody (Biotinylated) specific to Universal GSH. Excess conjugate and unbound available in test sample or standard are washed away, and Avidin-Horseradish Peroxidase (HRP) conjugate are added to each micro plate well and incubated. Next TMB substrate is added to each and every well The chemical reaction between enzyme-substrate is stopped by adding stop solution, due to which color of the solution turns yellow. In the last, the optical density (OD) of the wells are measured by spectrophotometer at a wavelength of 450 nm ? 2 nm. The concentration of Universal GSH in tested available in test samples can be calculated by comparing the OD of the available in test samples to the standard curve.
