Next TMB substrate is added to each and every well The chemical reaction between enzyme-substrate is stopped by adding stop solution, due to which color of the solution turns yellow. In the last, the optical density (OD) of the wells are measured by spectrophotometer at a wavelength of 450 nm ? 2 nm.
