This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate of the kit has been pre-coated with an antibody specific to Mouse ADP/Acrp30. Test samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a detection antibody (biotinylated) specific for Mouse ADP/Acrp30 and Avidin-Horseradish Peroxidase (HRP) conjugate are added to each well of the well and then the plate is incubated for specific time. The free conjugate is washed away by buffer. The substrate solution is added to each well. The well that contains Mouse ADP/Acrp30, detection antibody (biotinylated) and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is completed by the addition of stop solution and the color turns yellow. In the last, the optical density (OD) of the wells are measured by spectrophotometer at a wavelength of 450 nm ? 2 nm. The OD value comes in proportional to the concentration of Mouse ADP/Acrp30. The concentration is calculated for Mouse ADP/Acrp30 in the samples by comparing the OD of the samples with the standard curve.
