This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate of the kit has been pre-coated with an antibody specific to Rat PACAP. Test samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a detection antibody (biotinylated) specific for Rat PACAP and Avidin-Horseradish Peroxidase (HRP) conjugate are added to each well of the well and then the plate is incubated for specific time. The free conjugate is washed away by buffer. The substrate solution is added to each well. The well that contains Rat PACAP, detection antibody (biotinylated) and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is completed by the addition of stop solution and the color turns yellow. In the last, the optical density (OD) of the wells are measured by spectrophotometer at a wavelength of 450 nm ? 2 nm. The OD value comes in proportional to the concentration of Rat PACAP. The concentration is calculated for Rat PACAP in the samples by comparing the OD of the samples with the standard curve.
Tested Sample Types: serum, plasma and other biological fluids Assay Time: 3.5h Cross reactivity: This kit recognizes Rat PACAP in samples. No significant cross-reactivity or interference between Rat PACAP and analogues was observed Precision: Both intra-CV and inter-CV are < 10%.Storage and Expiry: 2-8 degree C./-20 degree C., 12 months
