Test Principle: This ELISA kit uses Sandwich-ELISA as the method. The Microelisa stripplate providedin this kit has been pre-coated with an antibody specific to mammalian target ofrapamyein,mTOR . Standards or samples are added to the appropriate Microelisa stripplatewells and combined to the specific antibody. Then a Horseradish Peroxidase (HRP)-conjugated antibody specific for mammalian target of rapamyein,mTOR is added to eachMicroelisa stripplate well and incubated. Free components are washed away. The TMBsubstrate solution is added to each well. Only those wells that contain mammalian target ofrapamyein,mTOR and HRP conjugated mammalian target of rapamyein,mTOR antibody willappear blue in color and then turn yellow after the addition of the stop solution. The opticaldensity (OD) is measured spectrophotometrically at a wavelength of 450 nm. The OD value isproportional to the concentration of mammalian target of rapamyein,mTOR. You cancalculate the concentration of mammalian target of rapamyein,mTOR in the samples bycomparing the OD of the samples to the standard curve.
